RESUMO

SHP2 has emerged as an important target for oncology small-molecule drug discovery. As a nonreceptor tyrosine phosphatase within the MAPK pathway, it has been shown to control cell growth, differentiation, and oncogenic transformation. We used structure-based design to find a novel class of potent and orally bioavailable SHP2 inhibitors. Our efforts led to the discovery of the 5-azaquinoxaline as a new core for developing this class of compounds. Optimization of the potency and properties of this scaffold generated compound 30, that exhibited potent in vitro SHP2 inhibition and showed excellent in vivo efficacy and pharmacokinetic profile.

Design rules and application spaces for closed-shell conjugated polymers have been well established in the field of organic electronics, but the emerging class of open-shell stable radicals has not been evaluated in such detail. Thus, establishing the underlying physical phenomena associated with the interactions between both classes of molecules is imperative for the effective utilization of these soft materials. Here, we establish that Förster Resonance Energy Transfer (FRET) is the dominant mechanism by which energy transfer occurs from a common conjugated polymer to various radical species using a combination of experimental and computational approaches. Specifically, we determined this fact by monitoring the fluorescence quenching of poly(3-hexylthiophene) (P3HT) in the presence of three radical species: (1) the galvinoxyl; (2) the 2-phenyl-4,4,5,5-tetramethylimidazoline-3-oxide-1-oxyl (PTIO); and (3) the 4-hydroxy-2,2,6,6-tetramethylpiperidine-1-oxyl (TEMPO) radicals. Both in solution and in the solid-state, the galvinoxyl and PTIO radicals showed quenching that was on par with that of a common fullerene electron-accepting derivative, due to the considerable overlap of their absorbance spectrum with the fluorescence spectrum of the P3HT species, which indicated that isoenergetic electronic transitions existed for both species. Conversely, TEMPO showed minimal quenching at similar concentrations due to the lack of such an overlap. Furthermore, computational studies demonstrated that FRET would occur at a significantly faster rate than other competing processes. These findings suggest that long-range energy transfer can be accomplished in applications when radicals that can act as FRET acceptors are utilized, forming a new design paradigm for future applications involving both closed- and open-shell soft materials.

Cartilage tissue engineering strategies often rely on hydrogels with fixed covalent crosslinks for chondrocyte encapsulation, yet the resulting material properties are largely elastic and can impede matrix deposition. To address this limitation, hydrazone crosslinked poly(ethylene glycol) hydrogels were formulated to achieve tunable viscoelastic properties and to study how chondrocyte proliferation and matrix deposition vary with the time-dependent material properties of covalent adaptable networks. Hydrazone equilibrium differences were leveraged to produce average stress relaxation times from hours (4.01 × 103 s) to months (2.78 × 106 s) by varying the percentage of alkyl-hydrazone (aHz) and benzyl-hydrazone (bHz) crosslinks. Swelling behavior and degradation associated with adaptability were characterized to quantify temporal network changes that can influence the behavior of encapsulated chondrocytes. After four weeks, mass swelling ratios varied from 36 ±â€¯3 to 17 ±â€¯0.4 and polymer retention ranged from 46 ±â€¯4% to 92 ±â€¯5%, with higher aHz content leading to loss of network connectivity with time. Hydrogels were formulated near the Flory-Stockmayer bHz percolation threshold (17% bHz) to investigate chondrocyte response to distinct levels of covalent architecture adaptability. Four weeks post-encapsulation, formulations with average relaxation times of 3 days (2.6 × 105s) revealed increased cellularity and an interconnected articular cartilage-specific matrix. Chondrocytes embedded in this adaptable formulation (22% bHz) deposited 190 ±â€¯30% more collagen and 140 ±â€¯20% more sulfated glycosaminoglycans compared to the 100% bHz control, which constrained matrix deposition to pericellular space. Collectively, these findings indicate that incorporating highly adaptable aHz crosslinks enhanced regenerative outcomes. However, connected networks containing more stable bHz bonds were required to achieve the highest quality neocartilaginous tissue. STATEMENT OF SIGNIFICANCE: Covalently crosslinked hydrogels provide robust mechanical support for cartilage tissue engineering applications in articulating joints. However, these materials traditionally demonstrate purely elastic responses to deformation despite the dynamic viscoelastic properties of native cartilage tissue. Here, we present hydrazone poly(ethylene glycol) hydrogels with tunable viscoelastic properties and study covalent adaptable networks for cartilage tissue engineering. Using hydrazone equilibrium and Flory-Stockmayer theory we identified average relaxation times leading to enhanced regenerative outcomes and showed that extracellular matrix deposition was biphasic as a function of the hydrazone covalent adaptability. We also showed that the incorporation of highly adaptable covalent crosslinks could improve cellularity of neotissue, but that a percolating network of more stable bonds was required to maintain scaffold integrity and form the highest quality neocartilaginous tissue.

RESUMO

Although less studied than their closed-shell counterparts, materials containing stable open-shell chemistries have played a key role in many energy storage and energy conversion devices. In particular, the oxidation-reduction (redox) properties of these stable radicals have made them a substantial contributor to the progress of organic batteries. Moreover, the use of radical-based materials in photovoltaic devices and thermoelectric systems has allowed for these emerging molecules to have impacts in the energy conversion realm. Additionally, the unique doublet states of radical-based materials provide access to otherwise inaccessible spin states in optoelectronic devices, offering many new opportunities for efficient usage of energy in light-emitting devices. Here, we review the current state of the art regarding the molecular design, synthesis, and application of stable radicals in these energy-related applications. Finally, we point to fundamental and applied arenas of future promise for these designer open-shell molecules, which have only just begun to be evaluated in full.

RESUMO

The psychological assessment of novice and experienced clerics is an important component of ordination, suitability, and risk evaluation to ensure that representatives of religious organizations are equipped, motivated, and safe for a life commitment to a faith vocation. It is the authors' opinion that such assessments should be conducted by skilled psychologists with expertise that covers occupational, clinical, and forensic domains. Further, the authors emphasize the importance of an objective and secular assessment to better inform the church about its role and responsibility for the oversight and spiritual development of the cleric. A thorough psychological assessment should incorporate a multimodal approach to information gathering, which includes a comprehensive review of background information and medical records, a personal history interview, a mental status examination, and administration of relevant psychometric measures and assessment tools. We also advise that, upon completion of the assessment, the requesting religious organization should be offered the opportunity to meet with the evaluating psychologist to discuss suitability issues and, if necessary, risk management planning.

Photoinduced charge-transfer (CT) processes play a key role in many systems, particularly those relevant to organic photovoltaics and photosynthesis. Advancing the understanding of CT processes calls for comparing their rates measured via state-of-the-art time-resolved interface-specific spectroscopic techniques with theoretical predictions based on first-principles molecular models. We measure charge-transfer rates across a boron subphthalocyanine chloride (SubPc)/C60 heterojunction, commonly used in organic photovoltaics, via heterodyne-detected time-resolved second-harmonic generation. We compare these results to theoretical predictions based on a Fermi's golden rule approach, with input parameters obtained using first-principles calculations for two different equilibrium geometries of a molecular donor-acceptor in a dielectric continuum model. The calculated rates (∼2 ps(-1)) overestimate the measured rates (∼0.1 ps(-1)), which is consistent with the expectation that the calculated rates represent an upper bound over the experimental ones. The comparison provides valuable understanding of how the structure of the electron donor-acceptor interface affects the CT kinetics in organic photovoltaic systems.

In organic photovoltaics many key ultrafast processes occur at the interface between electron donor and acceptor molecules. Traditional ultrafast spectroscopies, such as pump-probe or time-resolved fluorescence, are not ideal for studying the interface because most of their signal is from the bulk material. Time-resolved second-harmonic generation (TRSHG) spectroscopy solves this problem by only generating signal from the interface. We demonstrate an optically heterodyned TRSHG to reduce the impact of stray light, enhance sensitivity, and detect the full complex signal field.

Photosynthesis powers life on our planet. The basic photosynthetic architecture consists of antenna complexes that harvest solar energy and reaction centres that convert the energy into stable separated charge. In oxygenic photosynthesis, the initial charge separation occurs in the photosystem II reaction centre, the only known natural enzyme that uses solar energy to split water. Both energy transfer and charge separation in photosynthesis are rapid events with high quantum efficiencies. In recent nonlinear spectroscopic experiments, long-lived coherences have been observed in photosynthetic antenna complexes, and theoretical work suggests that they reflect underlying electronic-vibrational resonances, which may play a functional role in enhancing energy transfer. Here, we report the observation of coherent dynamics persisting on a picosecond timescale at 77 K in the photosystem II reaction centre using two-dimensional electronic spectroscopy. Supporting simulations suggest that the coherences are of a mixed electronic-vibrational (vibronic) nature and may enhance the rate of charge separation in oxygenic photosynthesis.

RESUMO

We demonstrate a "drop-in" modification of the pulse-shaped pump-probe geometry two-dimensional Fourier transform spectrometer that significantly improves its performance by making the measurement background-free. The modification uses a hybrid diffractive optic/pulse-shaping approach that combines the advantages of background-free detection with the precise timing and phase-cycling capabilities enabled by pulse-shaping. In addition, we present a simple new method for accurate phasing of optically heterodyned two-dimensional spectra. We demonstrate the high quality of data obtainable with this approach by reporting two-dimensional Fourier transform electronic spectra of chlorophyll a in glycerol/water at 77 K.

RESUMO

In many ultrafast contexts, a collinear pulse-shaping frequency-resolved optical gating (FROG) technique is desired. Some applicable techniques already exist, but they suffer from one of two issues: either they require many time points to allow for Fourier filtering, or they do not yield a traditional FROG trace. To overcome these issues, we propose and demonstrate a fast new phase-cycled FROG technique using a pulse shaper.

RESUMO

This article presents the first study on post-conviction polygraphy in the Netherlands. Importantly, it exclusively focuses on cybercrime offenders. The study is designed to systematically address the different child sexual behaviours exhibited by 25 participants who are in treatment for possessing child abuse images. The results indicate that post-conviction polygraphy can provide additional data to inform the development of theory in this area and contribute to the treatment, supervision, and more effective containment of offending behaviour and the reduction of future victimization.

This paper describes one of the first systematic focused attempts to elicit the views of sexual offenders about the treatment that they have received. The paucity of previous research in this area is suggested to be the result of negative societal attitudes towards sexual offenders, as exemplified by recent tabloid newspaper coverage of this population. It is suggested that these attitudes may have a negative impact upon the attitudes of clinicians and researchers, and thus upon the treatment offered to this client group. Therefore, it is argued that seeking the views of the clients is essential in order to ensure that treatment is relevant and reflexive, and promotes relapse prevention. A questionnaire was distributed to participants who had completed treatment groups run by the Psychology Service at a Regional Secure Unit and at a Probation Service Sexual Offender Unit. The questionnaire aimed to elicit their views of the group experience, including issues to do with programme structure and content, group process, and therapeutic style of group facilitators. The results (including a pilot study) indicated that, on the whole, participants' experiences of the group were positive, for example aspects of the group process, understanding and preventing reoffending. Some suggestions for the modification and development of future sexual offender treatment are outlined, based on the feedback from subjects, for example, consideration of alternative treatment modalities, enhancing the content of group sessions, improving the organization of groups and modifying the training of groups leaders.
